PhenoSense HIV Report Form

The PhenoSense™ HIV report is the final product of the sophisticated assay process. The results are depicted in a simple, easy-to-understand format that translates directly to treatment planning. There are two major components to the report, the fold-change bar graph and the drug susceptibility curves. Together, these two components build an accurate picture of drug resistance. The PhenoSense HIV report also includes Replication Capacity (RC) information for additional insight in treatment planning.

Phenotypic data —

Patient IC50:

The drug concentration required to inhibit replication of the patient virus by 50%.

Reference Range IC50:

The expected range of IC50 values for a drug-sensitive, "wild-type" virus. This value is determined by the IC50 value of a drug-sensitive reference virus tested with the patient virus.

Fold Change:

  • The fold change is the ratio of the Patient IC50 to the drug-sensitive Reference IC50. Patient IC50/Reference IC50 = Fold Change
  • Fold change 1.0 — patient virus exhibits the same degree of drug susceptibility as the drug-sensitive virus
  • Fold change less than 1.0 — patient virus is "More Susceptible" than the drug-sensitive reference virus
  • Fold change greater than 1.0 — patient virus is "Less Susceptible" than the drug-sensitive reference virus
  • Fold change equal to or greater than cutoff value — patient virus exhibits reduced susceptibility to that drug
  • Fold change less than cutoff value — patient virus is susceptible to that drug


Fold change bar graph —

The fold change bar graph is a visual representation of drug susceptibility that compares patient response to each drug to the response of the reference virus.
  • Blue bar appears in center box — patient virus exhibits susceptibility (fold change is less than the cutoff value)
  • Blue bar begins in center and extends left — patient virus is hypersusceptible or is more susceptible than drug-sensitive reference virus
  • Gray bar begins in center and extends right — patient virus exhibits reduced susceptibility


Drug susceptibility curve —

Data are analyzed by plotting the percent inhibition (y-axis) vs. log10 drug concentration (x-axis) for the patient virus and a drug-sensitive reference virus. Drug susceptibility curves are used to determine the drug concentration that is required to inhibit replication of the patient virus by 50% (IC50).
  1. Red curve = patient virus

  2. Blue curve = drug-sensitive reference virus

  3. Vertical and horizontal plot = identifies patient IC50

  4. Red curve same as or left of blue — patient virus is susceptible

  5. Red curve right of blue — patient virus is less susceptible
